بررسی مطالعه در مورد روش نگهداری سلولهای حیوانی در شرایط انجماد عمیق با تکیه بر دو ماده سوربیتول و ژلاتین و مقایسه آن با روش سرم و گلیسرول

Maintenance of animal cells in freezing conditions (cooling below zero degrees centigrade) is performed by special materials; these materials should protect the cell against temperature damages and the chemical and physical stress caused by freezing. The most common material used as Cryoprotectant (protective substances freezing) are Glycerol and DMSO ( - percent). This material are added to the base of a medium and serum and are frozen in liquid nitrogen, but due to various factors such as BSE agent (mad cow) and other infections related to the serum ،so serum-free Cryopreservation are used in new researches. To prevent the damages and protect the cells, in serum replacement the materials to be added to the freezing formula. The materials used for public cellular resistance is placed. Polyol and sugar are used for cellular resistance, cryoprotectant such as Sucrose, Trehalose, Sorbitol, Gelatin is used. Sugars have created a glass matrix molecule in cells that postpones molecular movement and decreases destructive reactions speed. In this study, substances such as sorbitol and sucrose and gelatin, and combinations of these materials were used and the results obtained compared with the positive control including samples containing with serum and glycerol and also the usual method of keeping the Theileriosis vaccine with % serum. After one year of maintenance and viability test of cells under different conditions showed that is significant differences in cell viability with cells stored in standard method and usual vaccine preservation method. Therefore, usual method used in the vaccine preparation that treated with % serum in preservation medium ، than forty years that are prepared in department of parasitic vaccines production, the best result shows besides the standard method.
