بررسی استانداردسازی پلاسمای خرگوش و خوکچه هندی تولیدی موسسه رازی

‭In research and diagnostic laboratories biological products such as hemolysin, plasma , serum , complement and defybrinated sheep blood are usually used. The research projects of standardization guinea pig and rabbit plasma , as a high consumption of biological products, was conducted in Razi Institute, dept.of laboratory animal science. In this study sampling was done based on SOP No. QCB.0313.SOP. From the three successive series of rabbits and guineapigs produced plasma , 8 vial, and a total 48 vials were sampled. (The plasma produced in accordance with the standards of laboratory animal breeding ) Parameters measuring protein , PH, hemoglobin and sterility test were determined. All samples in 2 ml deionized sterile water were desolved and divided into 200 macro-liter fractions and then labeled and stored at -20 C. The average protein concentration is 16/625 2/825 in Rabbit plasma and 15/042 2/136 in guinea pigs plasma. Hb concentration in rabbits plasma is 3/31 0/81 and in plasma of guinea pigs is 3/16 0/83. The result of cellulose acetate electrophoresis of rabbit and guinea pig plasma show that plasma samples related to rabbits contained six bands and guinea pigs contained five bands. The important point is that the pattern of guinea pig plasma proteins has a band less (group of protein) than rabbit plasma proteins pattern. Based on the results of densitometry of guinea pigs and rabbits plasma acetate electrophoresis, the average percentage of Alb, 1, 2, ‬ک‭, Fib and gamma in the plasma of the rabbits was 57/33 4/5, 9/14 1/3 , 7/16 1/02, 6/47 0/66, 13/44 2/76 and 7/02 1/29, and the mean percentage of Alb, a, ‬ک‭, Fib and gamma in plasma of guinea pigs was 50/84 4/43, 8/73 1/66, 12/2 2/75, 20/01 3/1 and 9/88 2/71. Results of pH measurement in plasma of rabbit and guinea pig indicate that the average pH were 8/42 0/15 and 8/05 0/1. The results show that coagulase test in rabbit plasma out of 24 samples one was negativ and of guinea pig samples two were negative. From sterility test results indicat lack of microbial contamination in all samples.‬
