papain digestion of beef meat (Linggood formula) is one of the media used for production of diphtheria toxin through the submerged culture method. I Although toxin produced by this method, in comparsion with Conventional route , show higher titer and potency as well as desirable economic concept ,however because of beef meat digestion source of ling good medium, this toxin may contain negligible amount of allergic antigens with the source of beef proteins . presence of allergen proteins in the final product leads to allergic reactions after repeated vaccination in humans. thus evaluation of final toxoid was conducted, from the toxin produced on Linggood medium ,for probable presence of beef sensitizing antigens before ues in vaccine composition was found to be necessary. In this research five experimental lot of Purified diphtheria toxoid, prepared by Linggood method in Razi institute , were studied using in vitro and in vivo tests. Dilutions 20,60 and 600 Lf/ml of each Lot of toxoid and related medium were evaluated in the primary stage by immunodiffusion test and then guinea pig sensitizing test (GPST) our observstion showed presence of beef sensitizing antigens in dilutions 60 and 600 Lf/ml of above mentioned toxoid and all the related mediums , but all of the 20 Lf/ml dilutions. except of one case showed abscence of sensitizing antigens.
