In vitro propagation of Damask Rose has played a very significant role in rapid multiplication of genotypes with appropriate traits and production of healthy and disease-free plants. In this study, we describe the effects of plant growth regulators on shoot proliferation of mature explants and root regeneration of two Iranian Rosa damascena accessions from nodal explants. The effect of different concentrations of BAP (1, 2, 5 mg/l) on number of shoots, shoot length (cm) and shoot dry weight (g) were studied in two Iranian accessions (Isfahan and Kerman) under MS medium. The shoots were transferred to a rooting regeneration medium, modified MS medium contained 1/2 concentrations ofmacronutrients and sucrose, full concentration of micronutrient supplemented with different concentrations of IAA containing 0.2, 0.5 and 1 mg/l. There was significant difference between the two accessions for number of shoots, shoot length and shoot dry weight. Number of shoots had higher values in Isfahan accession when compared with Kerman accession. The highest level of shoot multiplication rate (5) was recorded at a concentration of 2 mg/l BAP. The highest number of roots, root length and root dry weight were observed for 1 mg/l IAA. Therfore, genotype and growth regulators as key factors for in vitro propagation of R. damascena.
