Inhibitory effect of natural product Parthenolide on α-1-antiprotienase expression in breast cancer cell line

Introduction: Parthenolide is a Sesqiterpenlacton as major compound in Feverfew. Parthenolide has been predominately investigated as an anticancer agent numerous cancer types, both in vitro and in vivo. Apoptotic induction has been a new target mechanism for novel drug discovery. Precise mechanism that underlying PTL induced apoptosis is unclear. Method: In our previous study, we found this compound is an apoptosis inducer by flowsytometry, SEM microscopy and Annexin-V FITC test. In order to investigate the molecular mechanism of Parthenolideinduced apoptosis in MDA-MB231 cells, whole cell protein was extracted. Comparative proteome analyses of control and treated breast cancer cells were conducted after 24h trough 2-D analysis. Interesting spots were sent to York University (United Kingdom) for MALDI-TOF that related to control and treated protein of MDA-MB231 cells Result: Protein identification was obtained from NCBI and Uniport protein database. Among the proteins belonging to the signaling transduction category, α-1- antitrypsin, (spot 9), as anti- apoptosis protein was down regulated (5.1 fold) in treated cells with parthenolide. Conclusion: Our finding demonstrated that Parthenolid could reduce anti-apoptotic protein of α-1- antitrypsin. In this study we introduced a biomarker for early detection of breast cancer. Level of α-1- antitrypsin is elevating in this cancer and it decreased after treatment with Parthenolide
