چکیده مقاله:
Tuberclosis (TB) caused by mycobacterium tuberculosiswith almost 9 milion new cases each year remains of themost feared disease on the planet. The identification ofsources of infection, routes of transmission are importantpart of an eradication scheme and these can be achievedby differentiation of Mycobacterium tuberculosis isolatesby molecular typing method such as restriction fragmentlength polymorphism (RFLP). Golestan province afterSistan and baloochestan has the highest number of TBpatient in iran.in this study RFLP teqnique bytwo enzymespvu II and Alu 1 and PGRS probe were compared ofgenomic pattern diversity and better identify strain. FiveSputum speciement were collected from smear positivepatient in Golestan province. Samples were processedand culture on Lowenstein Jensen (LJ) slant for threeweeks. DNA was extracted from two sample based onvan embden protocol. First PCR assay targeting 16S rRNAgenes was used for identification of Mycobacterium spp.Whole DNA RFLP with Pvu II and Alu 1 restriction enzymeand hybridized with DIG labeled PGRS probe. In thisstudy, from 5 isolates, 2 different genomic patternwereobtained after digestion with Pvu II and hybridization withPGRS, and 3 different genomic patterns were obtainedafter digestion with Alu 1and hybridization with PGRS.
