Isolation and characterization of bacteriophage and assessment its antibacterial effects against clinical multi-drug resistant Pseudomonas aeruginosa strains

Background & Objective: Pseudomonas aeruginosa, sensitivity to antibiotic therapies has significantly decreased due to emergence of multidrug resistance strains. Using bacteriophages to fight antibiotic resistance strains is one of the alternative solutions. Herein, we aimed to isolate bacteriophages and investigate their antibacterial effect on clinical multi-drug resistant P. aeruginosa strains. Methods: Twenty-two clinical isolates of P. aeruginosa were obtained and characterized by biochemical and molecular assays. At first, antibiotic sensitivity pattern of collected strains to selected antibiotics was clarified via disk diffusion method then the strains with high antibiotic resistance were selected as a host for bacteriophage isolation. Bacteriophages were enriched and characterized by transmission electron microscope (TEM). Then the host range of bacteriophages was evaluated on the collected strains. Results: Two resistant strains (P۲ and P۳) and two susceptible strains (P۱ and P۹) to bacteriophages were selected for phylogenetic analysis using ۱۶S rRNA sequencing. Our results demonstrated that P. aeruginosa strains had lowest resistance to Colistin (۰%), Cefepime (۲۲.۷۲%), Ceftazidime (۲۲.۷۲%), and Meropenem (۳۶.۳۶%) while maximum resistance was belonged to, Piperacillin-tazobactam (۵۹%) gentamicin (۵۴.۵۴%), and amikacin (۵۴.۵۴%). TEM results indicated that bacteriophage PAP۲ belongs to the Caudouricetes class. Two bacteriophages, PAP۱ and PAP۲ were isolated from P۱ and P۱۶ strains and they exhibited similar and effective antibacterial action and host range. Conclusion: Our results revolve that bacteriophages PAP۱ and PAP۲ could inhibit the growth of P. aeruginosa by forming large transparent plaques and may serve as alternatives to antibiotics, however, further researches are recommended to guarantee the results.
