Identification and isolation of Lactobacillus acidophilus from traditional Doogh and its effect on Pseudomonas aeruginosa bacteria.

Lactobacilli are living microorganisms that, by settling in the intestinal environment, prevent the activity of unhelpful and pathogenic microorganisms. In addition, probiotics play an important role in maintaining the health of the consumer by synthesizing some essential substances. Lactobacillus acidophilus is one of the most useful species of Lactobacillus that is seen as a probiotic microorganism in dairy products. In this study, ۶۰ traditional dooghs were sampled. For this purpose, lactic acid bacteria were isolated by phenotypic methods and using a pair of specific primers for the ۱۶s rDNA gene, a specific band was created and the PCR product sequencing identified Lactobacillus acidophilus bacteria. Out of the total ۶۰ samples, ۳۸ cases of Lactobacillus acidophilus were identified. The inhibitory effect of the isolated lactic acid bacteria on Pseudomonas aeruginosa was investigated by the agar diffusion method using a disk. The diameter of the growth inhibition zone around the disk was measured. In order to reduce the error, each test was repeated at least three times and the average diameter of the growth inhibition zone on Mueller-Hinton agar medium was measured and recorded. The average diameter of the growth inhibition zone of Lactobacillus acidophilus on Pseudomonas aeruginosa was ۱۲ mm.
