Coronaviruses are a large family of viruses that can cause diseases such as severe acute respiratory syndromes. The most common clinical symptoms in SARS-CoV-۲ patients include fever, cough, fatigue, and gastrointestinal symptoms such as diarrhea and vomiting, although these latter symptoms occur less frequently. Considering that cell cycle disruption can lead to various immune system disorders, this study aims to examine changes in P۵۳ gene expression. One of the most effective therapeutic strategies is the use of Remdesivir, an adenosine nucleotide prodrug that becomes the active nucleoside triphosphate form once inside cells. This drug possesses inhibitory properties and, with comprehensive pharmacological results, shows promise as a treatment for this disease. This study aims to investigate the effects of Remdesivir on P۵۳ gene expression changes in COVID-۱۹ patients post-treatment. Blood samples from thirty COVID-۱۹ patients before and after Remdesivir treatment, as well as twenty healthy individuals, were collected for RNA extraction and cDNA synthesis. Following RNA extraction and cDNA synthesis, P۵۳ gene expression changes were assessed using Real-time PCR. Results indicated a ۵۰% reduction in P۵۳ gene expression in patients compared to the control group, which is statistically significant (P<۰.۰۰۰). Additionally, the treatment group showed approximately a ۱۰۰% increase in gene expression compared to the control group (P<۰.۰۰۰), also statistically significant. The difference in gene expression between the patient and treatment groups was found to be ۱۵۰%, statistically significant. It can be concluded that the coronavirus, by affecting genes related to the cell cycle, may cause complications such as an increase in white blood cell count in patients. Furthermore, Remdesivir, by increasing P۵۳ gene expression, significantly reduces the proliferation of immune cells, aiding in patient recovery.
