COVID-۱۹ emerged swiftly in ۲۰۱۹ and was declared a pandemic. Research indicates that the virus activates therapeutic pathways in patients suffering from severe illness. Consequently, it is essential to explore the pathways activated by this virus for effective COVID-۱۹ treatment. A significant inflammatory pathway involved in COVID-۱۹ is the JAK/STAT signaling pathway. Remdesivir, an antiviral medication, has been extensively researched for its efficacy against this disease. The objective of this study was to examine the variations in JAK gene expression, a crucial component of the activated inflammatory pathways, in COVID-۱۹ patients before and after Remdesivir treatment. In this study, blood samples were collected from ۳۰ patients infected with the Omicron variant of the coronavirus before they received Remdesivir (patient group) and after their final dose of the drug (treatment group). Additionally, samples were taken from ۲۰ healthy individuals as a control group. Following sample collection, RNA extraction and cDNA synthesis were conducted. The JAK gene expression changes were analyzed using real-time PCR, and statistical analysis was performed on the gene expression variations among the three groups. The results indicated that JAK gene expression in the patient group was ۵.۶ times higher compared to the control group (P<۰.۰۰۱). In the treatment group, Remdesivir successfully normalized the expression of this inflammatory gene. This reduction was statistically significant when compared to the patient group (P<۰.۰۰۱). The study's findings suggest that Remdesivir may have a beneficial effect on alleviating inflammatory conditions in COVID-۱۹ patients by lowering JAK gene expression.
