Investigating the effect of decreasing SLC۱۶A۱۳ gene expression on apoptosis and metastasis in Larynx cancer cell line

Eighty-five to ninety-five percent of oral cavity masses or tumors are malignant squamous cell carcinoma or squamous cell carcinoma (SCC), which is mostly seen in adults and often in the tongue and floor of the mouth. In addition, small interfering RNAs, by destabilizing the target mRNA, have a proven role in apoptosis or proliferation of human malignancies such as SCC. Meanwhile, SLC۱۶A, a member of the family of membrane transporters, which play an important role in hemostasis cells by using electrochemical potential difference or ionic gradient to transfer materials, has recently received attention due to its importance in the carcinogenesis process. The current study aimed to determine suppression of SLC۱۶A۱۳ gene expression on the ability of survival, growth and metastasis in laryngeal cancer cell line (Detroit ۵۶۲). First, Detroit ۵۶۲ cell lines were cultured in standard medium, and then synthetic siRNA was transfected to the target cells, with electroporation instructions, to reduce the expression of the target gene SLC۱۶A۱۳. The process of cell death and apoptosis was investigated by MTT test in triplicate repeats and flow cytometry. Scratch test was also used to investigate the role of this treatment on the migration ability of cancer cells. TRIzol was used to extract total RNA, followed by cDNA synthesis. Real Time PCR was used to analyze the effect of this treatment on target gene expression changes. Based on the results of this study, the decrease in SLC۱۶A۱۳ gene expression leads to an increase in the apoptosis and decreased viability of squamous cell carcinoma cells, but does not change the ability of cancer cells to migrate. Cell death in laryngeal cancer cells can be increased through the reduction of SLC۱۶A۱۳ expression, and the proliferation has been visibly reduced. Although the ability to prevent cell metastasis was not proven, it can be used as a therapeutic approach. However, more studies are needed to confirm this issue.
