Bioinformatic Analysis of Changes in TP۵۳ and hsa-miR-۱۲۲-۵p Expression and Their Interaction in Gastric Adenocarcinoma

The most common and lethal type of stomach cancers, which arise from glandular cells of the stomach, is gastric adenocarcinoma. Changes in TP۵۳ and hsa-miR-۱۲۲-۵p expression, and their interactions, were analyzed by using bioinformatics analysis in this study on gastric cancer. The TP۵۳ protein is among the tumor suppressor genes related to the cell cycle and response to DNA damage. Changes in the expression pattern of TP۵۳ may be associated with the onset of cancer. On the other hand, hsa-miR-۱۲۲-۵p is a microRNA involved in the regulation of some gene expressions. Results indicated that TP۵۳ was overexpressed, while has-miR-۱۲۲-۵p was downregulated in the cancer samples. The relation between TP۵۳ and hsa-miR-۱۲۲-۵p may point to the increased expression of TP۵۳ linked with the downregulation of hsa-miR-۱۲۲-۵p. The TargetScan analysis suggests that hsa-miR-۱۲۲-۵p may bind to the ۳' UTR region of TP۵۳ and modulate its expression regulation. It is indicated that the interaction of TP۵۳ with hsa-miR-۱۲۲-۵p may be considered a main regulatory mechanism; further experimental studies are needed to confirm these findings for an advanced understanding of its impact on TP۵۳ gene expression. TP۵۳ expression changes and hsa-miR-۱۲۲-۵p were gained from the TCGA data obtained from the UALCAN database in this study. One of the bioinformatics tools that has been used to predict how microRNAs interact with genes is TargetScan Human. Using this tool, we investigated the interaction of hsa-miR-۱۲۲-۵p with the ۳' UTR region of the TP۵۳ gene. by performing an appraisal with this tool, it was determined which target regions of the TP۵۳ gene are under the control of hsa-miR-۱۲۲-۵p. Results on the possible effects of this microRNA on TP۵۳ gene expression were investigated. In general, the results indicate that, although miR-۱۲۲-۵p interacts with TP۵۳, such interaction is not very influential in reducing TP۵۳ expressions. The investigation of such interactions might provide further insight into the molecular mechanisms involved in gastric cancer and even into the identification of new biomarkers since the TP۵۳ gene plays a major role in the disease and miRNAs play an important role in the regulation of gene expression. These interactions between TP۵۳ and hsa-miR-۱۲۲-۵p can thus be used as potential diagnostic and prognostic markers of gastric adenocarcinoma. High levels of TP۵۳ expression, combined with low levels of hsa-miR-۱۲۲-۵p, might give critical information on the condition of disease progression, tumor response to therapy, and diagnosis, aiding in the therapeutic approach in improving cancer.
