EPH Receptor B۱ gene expression is linked to the survival of PTC patients

Introduction Well-differentiated papillary thyroid carcinoma (PTC) is a frequently occurring type of thyroid cancer. The development of PTC is influenced by both environmental and genetic factors. Despite extensive research on genes associated with PTC, the exact genetic foundation of PTC is still not fully understood. Our study aimed to identify genes associated with PTC outcome. Methods An analysis was conducted on the GSE۱۰۴۰۰۵ dataset, which consisted of ۱۹ PTC tissues and ۵ normal tissues adjacent to PTC tumours. The validation of data quality required the application of log۲ transformation and quantile normalisation. A differential expression analysis was performed using the Limma program. Genes exhibiting logFC values greater than ۱.۲ and adjusted p values below ۰.۰۵ were selected as differentially expressed genes (DEGs). A total of ten samples of papillary thyroid carcinoma (PTC) and ten adjacent normal tissues were obtained after thyroidectomy surgery. We will conduct total RNA extraction, complementary DNA synthesis, and RT-qPCR. The relative expressions will be analysed using REST۲۰۰۹ analytical software. An analysis was conducted using RNAseq data to examine survivability, stage, and nodal metastasis. Results An analysis of microarray data revealed the discovery of ۱۸۰ differentially expressed genes (DEGs). One of the genes, EPH Receptor B۱ (EPHT۲), had a logFC value of -۲. The adjusted p-value is ۳.۴E-۰۳. An analysis of RNAseq data showed a noteworthy effect of EPHT۲ expression on decreasing the probability of patient survival (p-value of ۸.۷E-۰۳). EPHT۲ expression was assessed at different biological stages, showing a median expression level of ۱.۹۶ in Stage ۲ and ۱.۲۳ in Stage ۴. Our analysis found a significant association between the expression of EPHT۲ in Stages ۲ and ۴ and heightened aggression (P-value of ۱.۳۴E-۰۲). In tumours without infiltration into the regional lymph nodes (N۰), the median expression level of EPHT۲ was ۱.۷۹. However, in tumours with metastases to the axillary lymph nodes (N۱), it was ۱.۱۹. A comparison of EPHT۲ expression between N۰ and N۱ tumours showed a statistically significant difference (P-value = ۷.۹۹E-۰۴). There was no notable difference in the expression of EPHT۲ between male and female patients, as indicated by the statistical analysis. Conclusion Expression of the EPHT۲ gene was significantly lower in PTC compared to the normal tissues surrounding PTC tumours. The decreased expression of EPHT۲ was found to be correlated with improved patient survival and increased metastasis.
