Bioinformatic analysis of NFAT۵ gene expression changes in breast cancer

Cancer is the cause of death and reduced life expectancy in the world. Breast cancer is the most common malignant tumor in women and has the highest mortality rate; Its incidence and mortality are increasing and it tends to younger patients. Breast cancer is biologically complex and a heterogeneous cancer. In the eastern Mediterranean region, including Iran, breast cancer has the highest mortality rate among women compared to other cancers. Breast cancer accounts for ۱۲.۵% of all cancers in Iran, and it is the sixth cause of death in the country. The purpose of this bioinformatics study was to investigate NFAT۵ gene expression changes in breast cancer and introduce it as a diagnostic and prognostic biomarker. Materials and methods: In order to investigate changes in NFAT۵ gene expression in breast cancer, TCGA data provided by the oncodb database was used. (oncodb.org). Also, based on TCGA clinical data, the relationship between NFAT۵ gene expression and patient mortality rate and clinical characteristics was evaluated. Results: Our results showed that the expression level of this gene in cancer samples has decreased significantly compared to control samples. (LogFc = -۱.۳۶, FDR≤/۰۵). According to the study, the expression level of NFAT۵ in samples with TNM.T۴ status (Tumor, Node, Metastases) is significantly reduced in comparison with T۱ and T۲, and the expression level of NFAT۵ in samples with TNM.M۱ status in comparison with TNM.M۰ also has a significant decrease. In addition, K-M diagram showed that there is no significant correlation between NFAT۵ gene expression and survival characteristics of patients. (Logrankp=/۸۵). Conclusion: The results of the study showed that the expression level of NFAT۵ gene decreases in breast cancer and according to the obtained clinical information, it can be considered as a diagnostic biomarker in breast cancer. But more studies are needed to consider the NFAT۵ gene as a prognostic biomarker.
