Background: There are recently identified lncRNAs which aredifferentially expressed among active ulcerative colitis (UC),Dysregulated lncRNAs were mostly enriched in pathways thatregulate immune responses such as NF-κB pathway. In addition,lncRNAs also play an important role in the components ofthe intestinal barrier, including intestinal epithelial cells, macrophagesand gut microbiota. HOTAIR is a well-characterizedlncRNA that has been implicated in several diseases. While theimportance of rs۱۲۸۲۶۷۸۶ in the HOTAIR have not yet beenstudied in UC, our aim was to investigate HOTAIR gene singlenucleotide polymorphism (rs۱۲۸۲۶۷۸۶) and UC susceptibilityin Guilan province population.Materials and Methods: We explored the relationship betweenthe SNP and HOTAIR expression level in ۵۰ UC patients and۶۰ healthy individuals. The HOTAIR polymorphism was genotypedby tetra primer amplification refractory mutation systempolymerase chain reaction (ARMS-PCR). Quantitative measurementswere performed in triplicate and relative expressionwas measured using the comparative Ct method (۲−ΔΔCt). Tonormalize the expression of HOTAIR, GAPDH was used as aninternal control. Statistical analysis was conducted using Med-Calc and Graphpad Prism software. The difference of genotypedistribution of rs۱۲۸۲۶۷۸۶ between the case and controls wasof statistical significance (P =۰.۰۲۳).Results: The TT genotype was associated with a ۳.۸۵-fold increasedrisk (۹۵% CI =۱.۳۶-۱۰.۸۹; p=۰.۰۱) of UC as comparedwith the CC+CT genotype. The qRT-PCR analysis demonstratedthat the HOTAIR expression was significantly higher inUC patients compared to normal (p<۰.۰۰۰۱). The expression ofHOTAIR in TT homozygous was significantly higher than thatin CT and CC genotypes (p=۰.۰۳, p=۰.۰۰۷ respectively).Conclusion: In Conclusion: , the findings of the presentstudy revealed the importance of genetic variant of HOTAIR(rs۱۲۸۲۶۷۸۶) on UC susceptibility among the Guilan provincepopulation. However, further well-designed studies with largescalemulticenter investigations involving different ethnicities arerequired to verify the role of HOTAIR SNPs in UC susceptibility
