Investigation the protective effect of catechin nanosystems against aluminum chloride-induced oxidative stress

Catechins is a polyphenol with antioxidant properties, and nanoencapsulation can preserve its antioxidant properties and increase its bioavailability.This study aimed to investigate the protective effect of the catechin nanosystems against aluminum chloride-induced oxidative stress compared with catechin. For this purpose, catechin-loaded chitosan/alginate nanosystem was synthesized . Then, male Wistar rats aged range۱۲-۱۴ weeks were feded with catechin nanoparticles at a dose of ۱۰ mg/kg and catechin at a dose of ۵۰ mg/kg by gavage. At the end of the treatment period, the activity of antioxidant enzymes catalase (U/ml), glutathione peroxidase(U/ml), and glutathione redoctase(U/L) was measured in the serum sample. Catalase activity in the control , catechin, aluminum chloride , catechin nanosystem , aluminum plus catechin nanosystem and aluminum chloride plus catechin groups were ۳۹.۱۱±۲.۴۰, ۳۸.۶۸±۲.۹۴, ۱۹.۵۴±۲.۰۷,۳۹.۷۸±۵.۳۲, ۲۹.۳۵±۴.۶۵, and ۲۹.۰۷±۲.۵۷, respectively.The activity of glutathione peroxidase in control, catechin, aluminum chloride, nanosystem catechin, aluminum chloride plus catechin nanosystem, and aluminum chloride plus catechin groups were ۱.۶۹±۰.۱۵, ۱.۵۷±۰.۱۹, ۰.۷۸±۰.۱۳, ۱.۵۲±۰.۲۱, ۱.۲۹±۰.۲۴ and۱.۲۷±۰.۳۴, respectively.The activity of glutathione reductase enzyme in the control, catechin,aluminum chloride, catechin nanosystem,aluminum plus catechin nanosystem, aluminum chloride plus catechin were ۴۰.۹۲±۶.۴۱, ۳۹.۵۵±۲.۹۰, ۲۴.۹۰±۲.۴۷, ۳۸.۷۷±۲.۴۲, ۳۵.۲۳±۸.۹۲ and ۳۸.۶۶±۶.۰۶, respectively. As the results show, the activity of antioxidant enzymes in rate treated with aluminum chloride showed a significant decrease (p<۰.۰۰۱) in the control group, while the use of catechin or its nanoparticle prevents the reduction of enzyme activity. Since the lower dose of the catechin nanosystem was used, it is predicted that catechin encapsulation, caused the increased antioxidant property and reduced possible side effects of catechin.
