Study of anti-oxidative and anti-cancer effects of Rutin and Tamoxifen in the treatments of T-47D cell line

Tamoxifen is one of the most commonly used drugs in therapy of breast cancer but it has many side effects on various patients. In this research, the effect of simultaneous use of Rutin and Tamoxifen on T-47D cells and effects on the lethal concentration of Tamoxifen was investigated. Methods: Percent viability of T-47D cells as a breast cancer cell line was investigated in the presence of Tamoxifen, Rutin and their simultaneous use at 48 and 72 h of treatment using MTT assay. Results: The results showed that Tamoxifen, in the concentrations lower than 70 μM, produces ROS to kill T-47D cells. In upper concentrations, Tamoxifen can kill the cells via some other ways. In addition to the anti-oxidant effects, Rutin can trigger cell death in T-47D cells. Co-treatment of the cells with low concentrations of Rutin and Tamoxifen will be followed by fading anti-cancer effects of these two drugs because Rutin involves in neutralizing process of the Tamoxifen-produced ROS. Our investigations showed that for further inducing cell death, concentration of Tamoxifen should be decreased and the concentration of Rutin must be increased. Discussion: During the Tamoxifen-induced ROS production in the cells, Rutin molecules involve in neutralizing of ROS, so the toxic effects of Tamoxifen are diminished and Rutin cannot exert its anticancer effects by itself. In upper concentrations of Tamoxifen some other cellular mechanisms, which are different from the ROS production, are involved and the only way for maintaining of a distinct amount of cell death (like 40%) is to reduce the concentration of Tamoxifen and increase in the concentration of Rutin. So, in this way, side effects of Tamoxifen are attenuated and chemotherapy will be done more safely
