چکیده مقاله:

Globally Esophageal Cancer (EC) is the sixth leading cause of cancer mortality and the eighth common cancer. Iran is one of the regions with high rate of EC, which located on the geographic belt of EC. About 90% of EC belongs to Esophageal Squamous Cell Carcinoma (ESCC). The role of different types of noncoding RNA (ncRNAs) in cellular processes has received much attention. LncRNAs are one of the ncRNAs subsets that have a great role in the control of gene expression. Deregulation of lncRNA Often involve in cancer tumorigenesis, invasion and metastasis. LncRNA CCAT1 recentlydiscovered and located in 8q24 gene. This location was described as hub with much genetic alternation. Transcripts of lncRNA can affect the expression of various genes. According to the different studiesCCAT1 transcripts regulate the activities of some super-enhancers. Up-regulation of CCAT1 displayed in various types of cancer such as colon, gastric, and hepatocellular carcinoma. This investigationconducted on ESCC patients (n=40). The expression level of lncRNA CCAT1 evaluated tumor tissues and their adjacent normal tissues in EC patients. At first total RNA extracted from each sampleseparately. Then, cDNA was synthesized and expression level evaluated by quantitative Real Time PCR. The expression level of CCAT1 was normalized against those of GAPDH. the expression levelof CCAT1 in EC tumor tissue was higher than adjacent non tumor tissue. Discussion: this is the first report on expression changes of CCAT1 in EC. Evaluation of the expression of some lncRNA isprecious to find potential diagnostic and prognostic markers for cancer early diagnosis and treatment. This study demonstrated that up-regulation of lncRNA CCAT1 as an oncogene may be correlated withthis disease. Keywords: Esophageal Cancer, Esophageal Squamous Cell Carcinoma, LncRNAS, CCAT1.
