Breast cancer is the most common worldwide malignancy in females and it has been considering as a main reason for female morbidity and mortality. Genetic and environmental factors play an importantrole in relation to the incidence of breast. Several methods have been implied for the treatment depending on the stage of breast cancer including: mastectomy, chemotherapy, radiotherapy as well as chemoradiotherapy. Radiotherapy is one of the main and first steps of solid tumors which is conventionally is used as standard procedure after mastectomy. It is noteworthy that ionized radiationcan effect on marginally normal cells around tumor, so the probability of chromosomal abnormality will be increased in relation to emerging secondary tumor malignancy. Moreover, toxicity and poorprognosis regarding as other side effects of high dose radiation. Prediction of delayed toxicity induced by radiation in normal tissue will provide personalized radiotherapy in order to yielding better resultand therefore minimized the side effects. However, patients who show more tolerant are more susceptible for higher dose but who are more radiosensitive would not be exposed to conventional dose. therefore, other methods for treatment should be replaced. In this study 30 female patients have been recruited who were suffering from breast cancer and did not get any treatment. 5ml peripheral blood samples were collected in heparinized tube from each patient. After culturing peripheral blood lymphocytes for 72 hours at 37°C the cultures were exposed to 0.4 Gy X-radiation. After this period of time and preparation of the slides investigation of chromosomal and nuclear breakage were perform under the microscope field. Also this procedure was performed for 30 healthy controls and then the results were compared in these groups. The result of this study shows significant variation in chromosomal and nuclear breakage in radiosensitive and radioresistant patients in one hand and compare with the controls in the other hand. In conclusion the determination of patient’s radiosensitivity could lead us to choose optimal dose individually as a personalized medical treatment
