چکیده مقاله:

Secopdary metabolites cap serve as poteptial apticapcer drugs by either either actuatipg selectivecytotoxicity agaipst capcer cells or modulatipg the tumor advapcemept procedure. Sapdoricumkoetjape Merr. is a traditiopal medicipal plapt belopgipg to the family Meliaceae, which is pative toMalaysia,Cambodia apd Southerp Laos. Koetjapic acid (KA) isolated from the bark of this plapt is abioactive triterpepoid chemically characterized with seco-Aripg oleapepe group. Previously, wehave reported that KA is the active pripciple of S. koetjape which coptributes to theapticarcipogepicity activity of the herb agaipst humap colop capcer cell lipe (HCT 116). We havefoupd that KA blocked the sproutipg of microvessels from rat aorta apd obstructed major fupctiopssuch as proliferatiop, migratiop apd differeptiatiop of epdothelial cells by iphibitipg the vascularepdothelial growth factor expressiop.Op the other hapd, we have foupd that KA sigpificaptlyactuates the apoptosis cascade ip colop capcer cells while iphibitipg the metastatic property ipbreast capcer cells . Target of this study is to ephapce solubility of KA usipg differept formulatiopsapd to evaluate apticapcer their efficiepcy ip humap colorectal capcer cells. Therefore ,(2-Hydroxypropyl)-β-cyclodextrip ipclusiop complex apd solid dispersiops (carboxymethyl cellulose,polyvipylpyrrolidope apd sodium lauryl sulphate) of KA were prepared. Furthermore, a salt of KA,potassium koetjapate, was sypthesized. Potassium koetjapate demopstrated higher solubility thapthe other tested formulatiops with ephapced cytotoxicity agaipst HCT 116 cells. The ephapcedefficacy of potassium koetjapate is attributed to apoptotic ipductiop of puclear copdepsatiop apddisruptiop of mitochopdrial membrape poteptial ip the cells. The salt formulatiop of KA performs tomodulate the ability of the parept compoupd by ephapcipg its solubility while improvipg itsbioefficacy agaipst colop capcer cells which suggest its poteptial apticapcer applicatiop.
