چکیده مقاله:
Many pathological and physiological condition such as cancer, metastasis, aging and failureoxygen delivery can alter oxygen level in cell. Hypoxia is such a condition that oxygenconcentration decreases below its standard threshold. Indeed hypoxia triggers remarkablemolecular mechanisms to help cell adaption. Hypoxia inducible factor (HIF) is one of thewell studied factor which is induced in response to hypoxia and help to restore cellcondition by increasing available oxygen level, regulation metabolic state and elevate cellsurvival and proliferation. HIF complex is heterodimer protein composed of two subunitHIF1α and HIF1β. Although HIF1β is stable factor in cell, HIF1α’s stability is determinedby cell condition. In many cancers and solid tumor cells are in hypoxic condition, in thissituation HIF can promote cell condition and help to tumor progression. In fact high level ofHIF was seen in many cancers. One strategic way to control cells propagation is inhibitingHIF activity. In this way the amount of available oxygen decrease and cancer progressioncan be controlled. Control activity of HIF can be done in different situation from mRNAtranscription to DNA binding function of HIF. As stability of HIF is rate limiting part ofcontrolling its activity, any attempt to activate its degrading pathway can help to decreaseamount of HIF. Additionally another controlling mechanisms is preventing HIF activity byinhibit DNA-binding of this factor. Thus, many compound can provides a valuable chemicalprobe for inhibiting HIF activity in cancer cells. Furthermore, the work provides different smallmolecule inhibitor exerts potent anti tumor effects that targeting HIF
