چکیده مقاله:
According to the World Health Organization, breast cancer is the second most commoncancer among the women worldwide after skin cancer, threatening the life of thousandswomen each year. Breast cancer is the second leading cause of cancer death in women,exceeded only by lung cancer. Its rising incidence and mortality rates make it essential tounderstand the molecular mechanisms and affecting factors. Considering the conductedinvestigations, Metastasis is the main reason of breast cancer treatment failure that leads topatients death. Whereas researches indicate that microRNAs (which known as biologicalregulators for gene expression) play a critical role in initiation and progression of cancer,metastasis and resistance to various therapies, in the present study we comparing theexpression level of Mir-137 between primary tumor breast cancer tissue and metastaticbreast cancer tissue. In this study we investigated 60 breast cancer patients that hadmastectomy surgery and the sampled tissues were collected from 30 primary tumors and 30metastatic tumors from axillary lymph node then RNA extraction and cDNA synthesis wereperformed and finally the expression level change of this microRNA with Real-time PCRtechnique was explored. After analyzing the obtained data from performing comparative RealtimePCR method on the metastatic and primary tumor breast cancer patients, the expressionlevel of Mir-137 compared with reference gene (Mir-16) shows 7 times increasing in themetastatic breast cancer group. It is concluded that Mir-137 is introduced as a diagnosticbiomarker for recognition metastatic breast cancer patients and also a prognostic biomarker inprimary tumor breast cancer in order to early diagnosis of metastasis.
