Evaluation of Doxorubicin Inhibitory Effects in Myelogenous Leukemia Cells and Peripheral BloodMononuclear Cells Viability

چکیده مقاله:

Doxorubicin (also known as adriamycin) is the most commonly used anthracycline that hasintercalating binds effect which results reducing activity of Topo ІІα. It produces tumorshrinkage when used as therapy for a wide variety of cancers such as sarcomas, lymphomasand leukemia. It is used as part of combination chemotherapy regimens in leukemia.Doxorubicin has immunosuppressive effects and Production of white blood cells issuppressed after doxorubicin therapy. In the present study, we used to (ADRIAMCIN)® CSis produced by Pfizer in Australia, contain 2 mg doxorubicin hydrochloride /ml 0.9% salinesolution for intravenous injection and tested In Vitro condition. Number of (5×10) ^(5 ) K562cells (chronic myelogenous leukemia cell line) /ml, were pipetted into each well of 24-welltissue culture plate and were treated by various concentration of doxorubicin (50†400ng/ml) in cell culture standard condition. After 48 hours (IC)_50 value (half maximal inhibitoryconcentration) in K562 cell line was measured by MTT 3-(4, 5-dimethylthiazol-2-yl)-5-diphenyltetrazolium bromide assay. Non-adherent PBMC (Peripheral Blood MononuclearCells) were obtained from heparinized peripheral blood of three normal donors by (Ficoll-Paque PLUS)^™method (GE Healthcare instruction 71-7167-00 AG) and were pipetted intoeach well of 24-well plate (2×10)^6 cells/ml per well). Cells were treated up to four timesdilution of K562 doxorubicin (IC)_50 values (100†400 ng/ml) and cells viability after 48hours were measured by MTT assay. Values of doxorubicin (IC)_(50 )in K562 cells was 100± 3.2 ng/ml (Mean ± SE) but inhibitory effects of doxorubicin and viability in PBMC nosignificant differences were observed up to 400 ng/ml (P > 0.05), it seems that values of theinhibitory and cytotoxicity effects of the drug in non-adherent PBMC is high and the valueshad no effects on cells viability. Tests data were analyzed by using one-way ANOVAfollowed by the Tukey post-hoc test.
