Breast cancer is the second most common cancer globally and the leading cancer among women. While there are various therapeutic approaches available, there is an ongoing continuous demand for new natural products and resources to combat cancer. Among natural producing microorganisms, Actinomycetota (actinobacteria), a group of Gram-positive filamentous bacteria have been identified and used as an arsenal of biologics with antibacterial, antifungal, antiviral, immunosuppressive, and anticancer properties.
Streptomyces iranensis UTMC ۱۰۱ has been isolated from soil in Isfahan city, Iran. In this research, we analyzed its genome by
AntiSmash server and found ۱۴ anticancer genes. To evaluate anticancer activity of S. iranensis, it was cultured on ISP۲ agar as sporulation medium and incubated for ۱۴ days at ۲۸°C. Appropriate concentration of spore suspension was inoculated into the Erlenmeyer flasks containing ISP-۲ broth as seeding medium and incubated at ۲۸°C and ۱۸۰ rpm for ۴۸h. The seeding material (۱۰% v/v) was inoculated into the ۱۰۰۰ ml Erlenmeyer flasks containing ۳۰۰ ml fermentation medium and incubated at ۲۸°C and ۱۸۰ rpm for ۴۸h. The biomass was removed by centrifugation at ۲۵۰۰ rpm for ۲۰ min and the supernatant was extracted by equal volume of ethyl acetate. The organic phase was separated, and the solvent was evaporated at reduced pressure by rotary evaporator. The crude extract was weighed and appropriated serial dilutions (۰.۱-۳ mg/ml) were prepared in DMSO and its activity was assayed against (Michigan Cancer Foundation-۷)
MCF-۷ cell line. The cell line was cultured on DMEM supplemented with ۳?S Medium and incubated at ۳۷°C in an atmosphere of ۵% CO۲ to reach ۸۵% confluency. The treated cells were then incubated in a CO۲ incubator for three days, followed by flow cytometry analysis to evaluate the level of apoptosis. The IC۵۰ value determined from the MTT assay was ۱.۱۳ mg/ml, and flow cytometry indicated a significant level of apoptosis. In conclusion, we showed promising anticancer activity against the MCF۷ cell line. Further research is doing to extract and purify the chemical compound(s) having anticancer activity.
