Discovery of Novel microRNA and lncRNA Biomarkers for Prognosis and Diagnosis in Stomach Adenocarcinoma: An Integrative Bioinformatics and Systems Biology Approach

Stomach Adenocarcinoma(STAD) is the fifth most common cancer and the third leading cause of cancer-related deaths worldwide. Overall, ۲۶% of patients had single-site metastasis and ۱۳% had multiple-site metastasis. Complete tumor resection is the only way to treat this malignant disease. Since gastric cancer is usually asymptomatic in its early stages, many people's disease is diagnosed in its advanced stages, when the tumor is inoperable. Furthermore, because other conventional treatments (radiotherapy and chemotherapy) are only moderately effective for people with advanced or metastatic gastric cancer, the prognosis in such cases is poor. Recently, by conducting trials, promising results have been obtained regarding targeted molecular therapy, and the possibility has been strengthened that developing these agents can be a beneficial method. Material and methods: The Gene Expression Omnibus (GEO) database was utilized to find the dataset. The GSE۲۴۸۶۱۲ dataset was performed to detect genes that were expressed differently in STAD samples when compared to normal samples. Then the results of analyses were confirmed in The Gene Expression Profiling Interactive Analysis (GEPIA۲) and Analysis of RNA interactions (miRWalk and lncRRisearch) was conducted along with analysis of protein interactions (STRING). the significance level of microarray data analysis was considered ۰.۰۵ (adjusted p-value). Results: The experiment showed a notable increase in the levels of LINC۰۱۲۳۹ and NEAT۱ expression. NEAT۱ interacts with LINC۰۱۲۳۹ in the Stomach Adenocarcinoma samples analyzed, hsa-miR-۴۷۴۵-۳p had a increased expression level (Log FC= ۷.۷۱, adj P-value= ۲.۸۰e-۶). Conclusion: NEAT۱ and LINC۰۱۲۳۹ were found to be promising prognosis biomarker for Stomach Adenocarcinoma samples based on this computational.In the future, researchers will be able to gain more insight into the roles of NEAT۱ and LINC۰۱۲۳۹ play in Stomach Adenocarcinoma by investigating the correlation between NEAT۱ and LINC۰۱۲۳۹ and correlated lncRNAs, and microRNAs, especially the RNAs mentioned in this study.
