Antitumor activity of hydatid cyst fluid antigens on BALB/c mice with Breast Cancer

Hydatid cyst is the larval stage of Echinococcus granulosus, which leads to humoral and cellular immune responses in the host. We aim to investigate the effect of HCF and Antigen B (AgB) on ۴T۱ breast cancer cells in BALB/c mice. Methods: The livers of sheep infected with hydatid cysts were collected from Urmia slaughterhouse and the liquid inside the cyst was aspirated by a syringe and then concentrated using a centrifuge at ۲۵۰۰ rpm. AgB was obtained using a boiling water bath for ۱۵ min and centrifuged at Polyacrylamide gel electrophoresis for ۶۰ minutes with ۱۲.۵% sodium dodecyl sulfate (SDS PAGE) and Coomassie brilliant blue staining It was used to evaluate presence and quality. Mouse breast adenocarcinoma cell line (۴T۱) was cultured in rpmi medium and fetal bovine serum was added to it along with L-glutamine. After ۴۸ hours of incubation, the cells were harvested. ۴۰ mice were divided into ۴ groups, the first group was given PBS and the second, third, and fourth groups were injected intraperitoneally with concentrations of ۱۰۰, ۲۰۰, and ۳۰۰ μg/ml of HCF and AgB in ۱۰۰ μl of PBS along with naltrexone adjuvant. To determine the prevention effect of HCF and AgB antigens on ۴T۱ breast tumor Growth in BALB/c mice, ۱۰ days after cancer Cell inoculation, animals in each group In terms of tumor and tumor growth, blood was taken from one side of the mice and cytokines were measured based on immunological tests. Results: The results indicated that the mice that received different concentrations of the desired antigen along with the adjuvant had a significantly smaller tumor size compared to the control group, and the higher the injection dose, the smaller the tumor size. Was more Also, the level of Th۱ cytokines was increased compared to the control group. On the other hand, the level of Th۲ cytokines showed a significant decrease. Our histopathological studies showed a reduction in tumor volume and carcinogenesis. Antitumor activity of HCF and AgB may be related to immune responses against these antigens. Conclusion: Detailed attention is paid to the various antigenic components of helminth parasites are a suitable candidate for prevention and the treatment of cancers, autoimmune disorders, and inflammatory diseases because they control the immune system through regulators. This study evaluates the Antitumor activity of HCF and AgB on the ۴T۱ breast cancer cell line in BALB/c mice. Th۱ cytokines are involved in the upregulation of an effective anticancer immune response. Increased levels of Th۱ cytokines (IL-۲, IL-۱۲, TNF-α, and IFN-γ) in HCF and AgB groups can indicate that T cells are switching to a type I immune response.
