Introduction: Natural-derived Compounds, including marine organisms, are a favorable source for the production of drugs. Cassiopeia andromeda is called upside-down jellyfish one of the marine animals that utilize its venom with neurotoxin characteristics, hemolytic effect, antimicrobial activity, etc., which various studies indicate its properties against human cancers. miR-۱۸۱a-۵p is a microRNA with dual property that acts as an oncomiR in some human cancers that stimulate tumor growth. PTEN, as one of the targets of miR-۱۸۱a-۵p, is the most important inhibitor of the PI۳k/Akt signaling network, which is a key pathway in regulating the signaling of various biological processes such as metabolism, apoptosis, cell proliferation, and cell growth, and this pathway is important target for drug discovery with molecular effect. Methods: Jurkat cells were cultured next to peripheral blood lymphocyte cells (PBMC) as a control in RPMI-۱۶۴۰ medium and after that treated with IC۵۰ dose of venom, RNA was extracted and cDNA synthesis was performed, the relative expression level was measured by qPCR, and their significance was compared with Student's T-test and ANOVA. Results: The relative expression level of miR-۱۸۱a-۵p was reduced in Jurkat and PBMC cells, and the relative expression of both PI۳k and PTEN genes increased in PBMC while in Jurkat cells decreased. Conclusion: In this study, the effect of Cassiopeia andromeda venom in causing a change in the expression level of miR-۱۸۱a-۵p, PTEN, and PI۳k genes was seen, and this change in the direction of reducing the PI۳k/Akt pathway as an important signaling pathway in various types of cancers and also decreasing the expression of miR-۱۸۱a-۵p as an oncomiR, Ultimately, this study has shown that venom of C. andromeda can simultaneously suppress PI۳k pathway, and change the level of PTEN and miR-۱۸۱a-۵p so it is hoped that this toxin can be used as a drug in cancer treatment in the future.
