lncRNAs, a key regulator of the MAPK signaling pathway in gastric cancer

Gastric cancer is one of the leading causes of cancer related death worldwide. Various genetic and epigenetic factors are effective in the biogenesis and invasion of this lethal cancer. LncRNAs are one of the most important influencing genetic factors in the formation or prevention of various cancers, including gastric cancer. Therefore, we performed a systematic review to investigate the effects of lncRNAs on MAPK signaling pathway. Indeed, we aim to focus the attention on lncRNAs regulatory function on one of the most complex cellular pathways involved in GC proliferation, invasion, migration, and metastasis. Methods: The Google Scholar and PubMed database was systematically searched for trials in English language published between ۲۰۱۳ and ۲۰۲۴ that examined the negative or positive effects of various lncRNAs on MAPK signaling pathway in gastric cancer. Results: According to the conducted researches, a large number of lncRNAs effective on gastric cancer induce their effects through the control of the MAPK signaling pathway, and this control is basically induced by changing the expression of the main MAPK effector proteins such as ERK. This regulation of the pathway can occur as a direct effect on the expression of genes of the pathway or indirectly through miRNAs effective in the expression of proteins of the MAPK pathway. In this review, ۱۲ main lncRNAs affecting the MAPK pathway in gastric cancer have been examined along with their mechanism of action: LINC۰۰۴۸۳, LINC۰۰۱۵۲-۱, CASC۲, AOC۴P, CASC۱۹, MALAT۱, BCAR۴, PICART۱, HULC, H۱۹, SNHG۶ and GAPLINC. Conclusion: lncRNAs are known as molecules that are directly or indirectly effective in various cancers, including gastric cancer. The available evidence supports the role of this lncRNAs as an inducer, enhancer or inhibitor genetic factors in gastric cancer. Also, the advances on mechanism of understanding lncRNAs in MAPK signaling might bring novel candidates as biomarkers and therapeutics for GC.
