Background: This study aims to examine and determine the correlation between the drug Ciprofloxacin and breast cancer using datasets from PubMed and ScienceDirect.com. Ciprofloxacin is one of the most widely used drugs worldwide, and numerous studies have investigated the relationship between Ciprofloxacin and breast cancer. In addition, ۹۰% of mortality is due to cancer invasion and metastasis . Thus, early detection of cancer and the identification of anti-cancer agents with promising anti-metastatic potential are important cancer therapy. However, the results have been inconsistent. To obtain a more precise estimate, a systematic analysis of the relevant population-based studies was conducted. Materials and methods: In this review study, a search was conducted in the databases PubMed and Web of Science using the keywords (breast cancer) and (Ciprofloxacin) for the period between ۲۰۲۰ and ۲۰۲۴. The initial search yielded ۲۰ articles. Following an initial review and assessment of the articles characteristics and accessibility, ۶ studies were selected for further examination. Results: Ciprofloxacin inhibited the proliferation, colony formation, and migratory capabilities of both cancer cell lines. Furthermore, it induced apoptosis and caused cell cycle arrest in the G۲/M phase in MCF-۷ cell lines and in the S phase in MDA-MB-۲۳۱ cell lines.In addition, it up-regulated the expression of pro-apoptotic factors, p۵۳, PUMA and NOXA, and down-regulated the expression of anti-apoptotic factors, MDM۲ and MDM۴. At the same time, it inhibited epithelial-mesenchymal transition by increasing the expression of E-cadherin and decreasing the expression of TGF-B۱, SNAI۱, TWIST۱, MMP۲, and MMP۹. This FDA-approved drug (Ciprofloxacin) may potentially be applicable in the treatment of bladder cancer. Conclusion: The novel chalcone Ciprofloxacin demonstrated promising apoptotic and anti-metastatic activities against the MCF-۷ and MDA-MB-۲۳۱ breast cancer cell lines. Therefore, Ciprofloxacin is an attractive molecule for drug development in the treatment of breast cancer. Further studies of this new chalcone are required to increase our understanding of the molecular mechanisms underlying its effects.
