Introduction: This study aims to identify the main co-expressedgenes in the diagnosis and discover their mechanism ofaction in the occurrence and progression of diabetic retinopathyusing the RNA-Seq method. Therefore, we first collected datafrom the only RNA-seq expression profile of RD patients andthen performed the necessary bioinformatic analyses on them.Methods: In the above analysis, a total of ۵۸,۰۰۰ genes wereexamined, which reached ۳۸,۰۰۰ in the initial stages of filtering,and ۱۸,۰۰۰ genes after removing duplicate genes, whichwere then divided into Macula and Periphery groups. Examina tion of DEGs in two groups, Macula and Periphery, identifiedgenes with the highest differential expression separately, andtheir results are given.Results: Comparing these data with the results obtained froma study conducted on the same RNA-Seq profile based onGSE۱۶۰۳۱۰, in total, a list of candidate genes that probablyplay a role in the occurrence and progression of RD diseasewas introduced, and these results were then compared with thedata obtained from the pathways enrichment analysis showedthat DEGs are significantly involved in the biological processesof metabolic processes, and this can be an explanation of themechanism of the retina's response to environmental pressuresand metabolic stresses.Conclusion: The analysis of all cellular RNAs based onGSE۱۶۰۳۰۶ and the cellular miRNA data based on GSE۱۶۰۳۰۸were classified, and finally, by integrating the results of thesestudies, a list of miRNAs were introduced among all the expressedmiRNAs that showed a significant association with theproposed target genes related to the disease. Finally, a list of ۴miRNAs whose targets were included in the group of diseaseassociatedgenes and showed the most significant negative correlationwith each other was introduced as proposed candidatesof biomarkers for this project
