Background: After liver cancer and lung cancer, gastric cancer(GC) is the third most common cause of cancer-related mortality.It is one of the most dangerous malignant gastrointestinalneoplasms globally. According to the most recent data fromthe GLOBOCAN database, in ۲۰۲۰, there were over ۷۶۹,۰۰۰fatalities and over a million new cases of GC. Numerous biomarkerslinked to DNA, RNA, exosomes, etc. have been foundthanks to recent advances in genome analysis. Long non-codingRNAs (lncRNAs) appear to have a vital role in many cancers,according to recent studies. LncRNAs may be used as potentialbiomarkers to forecast the presence and prognosis of GCbecause of their high stability. Therefore, the purpose of thisinvestigation was to find out if LncRNA MMP۲۵-AS۱ might bea new diagnostic biomarker for GC.Materials and Methods: In this work, bioinformatics analysiswas used to look into the status of several genes' expressionin GC patients (microarray data analysis downloaded from theGEO database). Then, using the ENCORI database, the expressionof MMP۲۵-AS۱ was calculated bioinformatically. Followingsample collection, total RNA was isolated from ۲۰ GC tissuesamples and ۲۰ adjacent healthy tissue samples using anRNA extraction kit. Real-time PCR was used to assess the levelof lncRNA MMP۲۵-AS۱ expression in all samples followingcDNA synthesis.Results: Potential GC biomarkers discovered through the bioinformaticsstudies were tested for expression using a real-timePCR technique. An impressive dysregulation of the lncRNAMMP۲۵-AS۱ was found in the GC samples, according to bioinformaticsand experimental investigations.Conclusion: lncRNA MMP۲۵-AS۱ may be a diagnostic biomarkerthat is dysregulated in GC patients.
