Background: colorectal cancer is a top cause of death globally.Chemotherapy leads to side effects, urging the need fornovel therapeutic approaches. RNA-based therapy's versatilityin modulating numerous targets draws significant interestSuccessful outcomes in treating CRC by using nanoparticles toaim at several molecules like siRNA have been observed. NPstargeting siRNA can silence oncogenes and MDR genes. Wediscuss siRNAs targeting NP for CRC treatment, highlightingRNA-based therapy benefits of specificity, potency, and lowtoxicity compared to traditional treatments. We review recentRNA-based therapy and CRC clinical research, benefits, andpotential challenges to treating human CRC.Materials and Methods: This article reviews siRNA-basedtherapies, including target selection, potent siRNA sequences,and optimal delivery methods through nanoparticles. Theseconsiderations serve as a guide for developing novel siRNAtreatments via nanoparticles. we select articles based on inclusioncriteria covering nanoparticles in CRC treatment ranging۲۰۱۰-۲۰۲۰ in scientific databases like Medline, Elsevier, IEEEand Science direct.Results: Our findings demonstrate the viability of nanoparticlesas a candidate vector for siRNA-based colon cancer genetherapy. The results however imply that it is still crucial to developnew siRNA delivery vectors that are appropriate. Thestrong adsorption ability to organisms, few side effects, abundance,and low cost of polymer-made nanoparticles as transfectionvectors have made them successful as a drug or geneticcarrier in recent years.Conclusion: A deeper comprehension of the RNAi mechanismwill be the driving force behind even more comprehensivemethods for siRNA sequence selection and modification. Thespecificity and effectiveness of siRNA therapeutics will be enhancedby ongoing developments in nanoparticles utilization
