چکیده مقاله:
Background: Breast cancer is an extremely heterogeneousdisease and is dependent on various factors. The expression ofnumerous transcription factors that are involved in pH dysregulationis a major concern in solid cancers. This study aimed toinvestigate the effects of glucose and hypoxia conditions ongene expression of E۲F۱ and C/EBPA transcription factors inMCF-۷ and MDA-MB-۲۳۱ breast cancer cells.Method and Material: The breast cancer cells were culturedin presence of different concentrations of glucose and hypoxia.Cobalt chloride created hypoxia conditions at a concentrationof ۱۵۰ μM in culture media. Finally, mRNA expression levelsof C/EBPA and E۲F۱ transcription factors were investigated byReal-Time PCR.Results: The results showed that cell viability and proliferationof both cell lines are associated with glucose concentration aswell as hypoxia conditions. The expression of both C/EBPA andE۲F۱ genes indicated a significant correlation with different glucose and oxygen levels concentrations in breast cancer cells.
