چکیده مقاله:

Background: Childhood Acute lymphocytic (lymphoblastic)leukemia (ALL), which is caused by the clonal proliferation ofimmature T- or B-lymphoid cells in the bone marrow, is themost common hematologic disorder in childhood, accountingfor approximately ۳۵% of all childhood malignancies. The ceRNAshypothesis describes a domain of long non-coding RNAs(lncRNAs), pseudogenes, circular RNAs (circRNAs) and mRNAsthat compete with and bind to microRNAs (miRNA) andregulate their activity. In recent years, many studies of the networkceRNAs have been reported in the progress of the studyand treatment of leukemia types. Therefore, considering thepotential power of lncRNAs and miRNAs in the diagnosis andprognosis of ALL disease, in this study we will identify the biomarkersinvolved in this disease through bioinformatics analysisand Real Time PCR technique.Materials and Methods: By performing bioinformatics analysisthrough the weighted gene co-expression network analysis(WGCNA) package in R software, the co-expression genes thatact the same in terms of biological processes or pathways arefound in a common co-expression network, based on the type of network and the correlation of the co-expression genes inthe module were grouped together. In the following, using RealTime PCR technique, the expression of two key genes obtainedin ۵۰ people with ALL was investigated in comparison with thesame amount of control samples.Results: Bioinformatics analysis performed in this researchidentified biomarkers involved in ALL disease. RMRP as lncRNA,and H۲AC۱۷, as mRNA identified with the highest rate ofgene significant with increased expression.Conclusion: Considering the identified biomarker role of lncRNA,miRNA, and mRNAs in the development and progressionof patients with ALL and other cancers, it is suggested In orderto improve the treatment and prognosis of the disease and increasethe overall survival rate, more studies and investigationsshould be conducted in this field.
