Background: Colorectal cancer (CRC) is the third most commonlydiagnosed cancer worldwide (۱,۲), and its incidence isannually increasing (۳,۴). Various factors are associated withCRC progression, including genetic makeup, age, sex, ethnicity,inappropriate lifestyle (۵). This study aimed to identify thedifferentially expressed genes in the HCT۱۱۶ and DLD۱ celllines for progression of colon cancer.Materials and Methods: The GSE۵۸۹۰۴ accession numberwas selected from GEO datasets to identify the differentiallyexpressed genes between HCT۱۱۶ and DLD۱ cell lines. TheGEO۲R tool was performed to normalize and find differentiallyexpressed genes (DEGs) between compared groups. AdjustedP-value > ۰.۰۱ and log۲FC > ۱ were considered as statisticalsignificances. Protein-Protein Interaction was performed usingthe String database.Results: Based on the results, in the HCT۱۱۶ cell line comparedto DLD۱ cell line, upregulated genes include: GTSF۱, HBE۱,OR۵۱B۴, DKK۱, CYP۲۴A۱, ANO۶ and RAB۳۱ and downregulatedgenes include: OLR۱, TM۴SF۱, AGR۲, TSPAN۸,LGR۵, TMEM۵۲B and HIST۱H۳I.Conclusion: The bioinformatics and systems biology methodswere used to find out the important genes related to proliferation,differentiation and apoptosis in HCT۱۱۶ and DLD۱ colorectalcancer cell lines. These methods based on computationaltechniques and clinical validation is required
