Background: Biomarkers that can facilitate disease detection,staging, and projection of outcomes are highly desirable to enhancesurvival and to help determine optimized treatment forcolorectal cancer patients. MicroRNAs (miRNAs) are smallnon-coding RNAs that play a crucial role in gene regulatorynetworks(۱). The deregulation of miRNA expression has beenobserved in various types of cancer and may represent a novelclass of cancer biomarkers. Studies have demonstrated thatmiR-۱۳۵b is significantly expressed in tumor samples comparedto standard ones. MiR-۱۳۵b was differentially expressedin stage III colorectal tumors(۲).Material and Materials and Methods: In this project, aftermiRNA target gene prediction by the miRWalk database, weanalyzed TCGA RNA-seq data and predicted gene expressionpatterns(۳,۴).Results: The results of miRWalk analysis show the strong interactionof miR-۱۳۵b with PDLIM۵, and the findings of colorectalcancer genomics data analysis show a significant drop inthe expression of PDLIM۵ in the tumor sample compared to thestandard one. The reduction in miR-۱۳۵b expression in stage IIIpatients is also higher than in primary tumor samples.Conclusion: According to the location of miR-۱۳۵b bindingto PDLIM۵ and this negative relationship between increasedmiR-۱۳۵b expression in tumor samples and decreased PDLIM۵expression in tumor samples, regulation of PDLIM۵ expressionby miR-۱۳۵b can be predicted as well as decreased PDLIM۵expression in stage III patients and increased miR-۱۳۵b expressionas well. In addition, according to the findings of the TCGAwebsite, it is predicted that the reduction in gene expressionplays a role in the progression of cancer to a higher stage andthe growth of primary tumors.
