Background: PI۳Ks are a group of lipid kinases that phosphorylatethe ۳-hydroxyl group in the inositol ring of phosphatidyl-inositol molecules. Considering the very importantrole of the PI۳K/Akt signaling pathway in the cell, it is notsurprising that mutations in these kinases cause many diseases,cancer in particular. About ۴۰% of ER+/HER۲- breastcarcinomas have mutations in the PIK۳CA gene. One of theseclinically important mutations is H۱۰۴۷R, which occurs inexon ۲۰ of this gene.Methods and Results: First, the H۱۰۴۷R SNP was chosen to be produced in MCF۷ cell line by the utilization of CRISPR-Cas۹technology. Afterward, the subsequent tests were performed todeclare that producing isogenic mutants had been successful.Eventually, a sequencing analysis confirmed this claim. Successively,in order to analyze the effect of this mutation on theactivity of the PI۳K/Akt signaling pathway, western blot analysiswas accomplished.Conclusion: Identifying the effect of different mutations of thePIK۳CA gene involved in ER+/HER۲- breast cancer is a crucialstep to achieving an efficient pharmaceutical administration.With the isogenic mutants being available, it becomes possibleto conduct many intelligent and new Scientific studies
