P۲۳ protein is a candidate for vaccination against Cryptosporidiosis

Cryptosporidiosis is an important parasite causing severe diseases in the immunodeficient people especiallyAIDS patients. Cryptosporidiosis has been also reported as a common serious primary cause of outbreaks ofdiarrhea in newborn calves. The aim of this study was to analyze different bioinformatics’ characterizationof sporozoite surface antigen P ۲۳ of Cryptosporidium parvum.Our result showed that this protein has ۱۱۱ amino acids, MW ۱۱۲۳۲.۳۹ Da and PI ۵.۱۸. the number ofnegatively and positive charged residues was ۱۶ and ۱۴ residues, respectively. The total number of atomswas ۱۵۶۴. The estimated half-life for P ۲۳ protein was as follows: ۳۰ h (mammalian reticulocytes, in vitro),> ۲۰ h (yeast, in vivo) and > ۱۰ h (Escherichia coli, in vivo). The instability index (II) is computed to be۷۰.۱۱ and Aliphatic index was ۴۴.۵۹.Also, our survey showed that P ۲۳ has high antigenicity and it is not an allergen. This protein has four Serinphosphorylation sites and one acylation site. Transmembrane domains and subcellular localization of P ۲۳were ۶۲.۲ %: nuclear, ۸.۷ %: cytoskeletal, ۴.۳ %: cytoplasmic and ۴.۳ % plasma membrane. Secondarystructure prediction in the P ۲۳ sequence showed to ۵۱ (۴۵.۹۵%) alpha helix, ۵۸ (۵۲.۲۵%) random coil and۲ (۱.۸%) extended strands. moreover, ۳D structure of this protein was predicted and refined. For this protein,three high-ranked linear epitopes were determined using BCPREDS server. Also, at least five linear epitopeswith high score were confirmed by ABCpred Prediction Server.
