Liver tissue transcriptome data analysis for identification of key miRNAs in sheep (Ovis aries) with different dites

Background: Today, genetic engineering methods used to change the genome in a way to produce moreproducts like milk. Understanding regulatory mechanisms and the identification of small RNAs involved inmetabolism is one of the most challenging problems in genetic science. Therefore, the aim of the presentstudy was to identify some regulator genes in Ovis aries with different diets using bioinformatics analysis.Materials and Methods: In this research, ۱۳ samples of Illumina-sequence transcriptome data for sheep(Ovis aries) livers with different diets (high energy group n=۳, oil supplement group n=۳, light grazing n=۲,over-grazing n=۲ and control samples n=۳) were collected. Firstly, quality control of data were done withFastQC program. Then, the reads aligned to the reference genome (ARS-UI_Ramb_v۲.۰) with STARsoftware. Finally, to identify the differentially expressed miRNAs, data were analyzed using FeatureCountsand DESeq۲ programs, respectively.Result: Our results showed several differentially expressed miRNAs in each group (high energy group n=۱۳,oil supplement group n=۱۱, light grazing n=۱۰ and overgrazing n=۱۰). For example, in oil diet, miRNA۲۲۱and miRNA۲۹B significantly upregulated (p<۰.۰۵) that related to glucose metabolism. Also, the resultshowed expression of miRNA۳۰ increased in oil supplement groups which has a key role in lipid metabolism.In the high energy and overgrazing groups, miRNA۱۴۳ was significantly downregulated (p<۰.۰۵). Studiesshowed that this microRNA is an important regulator in glucose metabolism. In addition to, the resultedshowed that the expression of mir۱۶b, mir۳۰b, and mir۱۹۹a significantly decreased (p<۰.۰۵) in all groups.Studies indicated that these miRNAs have the important roles in glucose methbolism and immune systemresponse.Conclusion: In this study, our results indicated that different diets are determinant in regulating of keypathways and mechanisms like lipid metabolism and miRNAs can be candidate targets for purposefulnutrition.
