Bioinformatics study of level expression of miR-۲۱۲ as a biomarker of Glioblastoma

Introduction: MicroRNAs are small non-coding nucleic acids (around ۲۰ nucleotides) that are able to posttranscriptionallyregulate gene expression by binding to complementary sequences of target messenger RNA.In this study, miR-۲۱۲ expression levels, target genes and genetic pathways in glioblastoma were investigatedusing a bioinformatics database.Methods: This bioinformatic study was performed by several main databases, such as "miRdSNP, mirbase,GeneCards and DAVID". At first from mirbase, the chromosomal profile of this miRNA was obtained. Theposition of miR-۲۱۲ on the human chromosome and complete information about that, was identified InGeneCards. Finally the DAVID database was used for genetic pathways evaluation.Results and Discission: MiR-۲۱۲ gene is located on chromosome ۱۷p۱۳.۳. miR-۲۱۲ is overexpressed incancers of the central nervous system and is found in various parts of the cell, particularly in the extracellularspace and plasma membrane. miR-۲۱۲ plays a role in the post-transcriptional pathway of ۲۹۵۰ genes ofcellular sections. In addition, of the ۲۵۶۲ identified miR-۲۱۲ gene targets, which with influence on thesegenes could result in a variety of diseases, it can act as a tumor suppressor in glioblastoma by effecting on ۵۳genes. It was also found that miR-۲۱۲ by Suppression of SGK۳ gene as an important tumor-promoting geneis directly involved in the suppression of glioblastoma. Based on these results, miR-۲۱۲ inhibits glioblastomacell proliferation.Conclusion: The current study shows that, increased expression of miR-۲۱۲ in brain cells as a tumorsuppressor, can indicate the presence of cerebral glioblastoma, which may also lead to a rapid diagnosis ofthis chronic disease. Especially, since miR-۲۱۲ can also be detected in serum and plasma, which are muchmore readily obtainable than tissues, it attracts increased clinical attention as a biomarker in glioblastoma.
