Objectives: Because of abundant antigenic shift in influenza virus, most vaccines cannot perfectly protect against disease.The influenza viruses have some conserved epitopes like M۲e and HA۲ that are good candidates for production of universal vaccines.The matrix protein ۲ ectodomain (M۲e) is۲۳ amino acid residues in length and is highly conserved among influenza A viruses. M۲e-specific immune responses elicited by seasonal vaccines or natural infection are very weak. Nevertheless, vaccines that comprise M۲e fused to a carrier can induce strong M۲e-specific immune responsesand confer protection against various influenza A viruses. The aim of this study was to assess humoral immune response of broiler chickens after injection of M۲e (۲-۹)-gVIII. Materials & Methods: The M۲e (۲-۹)-gVIII gene cloned into PIT۲ expression vector. After expression and purification of the hybrid phage, SPF broiler chickens were immunized two times with three weeks’ interval with M۲e (۲-۹)-gVIII (hybrid phage). After the second immunization,final bleeding was performed and anti- M۲e (۲-۹)-gVIII IgY was measured by ELISA. Results & Conclusion: The results of ELISA showed that the level of anti-M۲e (۲-۹) peptide IgY antibodies washighly increased in immunized chickens. This hybrid phage is a potential candidate for production of antibodies that could recognize a broad range of subtypes of Influenza A virus and probably for vaccination.
