Diphenylene Iodonium (DPI), a NADPH oxidase inhibitor, improved aluminum phosphide poisoning: An Experimental Study

Background and objective: Diphenylene iodonium (DPI), a potent inhibitor of NADPH oxidase, is thought to significantly suppress Reactive oxygen species (ROS) levels. It was reputed to serve as one of the direct scavengers of ROS, and has ability to arrest completely the oxidative condition. Screening of the reports lead us to conclude that mitigation properties of this potential molecule are yet to be explored against Aluminum phosphide poisoning exerted stress condition. The aim of this study was to investigate the effect of DPI on aluminum phosphide poisoning-induced oxidative stress in an animal model. Materials and Method: : Male Wistar rats (12 weeks old) were divided into four equal groups (n=6) and treated as follows: control treated with saline, CDPI group received single dose of DPI (1 mg/kg. ip), ALP exposed with ALP (10mg/kg, orally) and DPI injected DPI (1 mg/kg. ip, 30 min later) in Alp exposed rats . Finally 24 hours after of drug administration, rats were sacrificed, and serum was extracted and evaluated for Oxidative stress parameters. The liver and heart tissues also were evaluated for oxidative stress indices. Additionally the fatality rate was recorded in each group. Findings: no death occurred in control groups. Alp toxicity was fatal in about 44%, DPI administration was effectively reduced it to 0%. The mean concentration of plasma malondialdehyde (MDA),Glutathione peroxidase (GPx) and Catalase activity significantly decreased in DPI treatment group compared to AlP group (p<0.05). Significant increase in MDA level of liver tissue in ALP group compared to control group was observed. The mean GPx activity in CDPI group were lower than ALP group and in AlP group were lower than control group. The mean catalase level of liver in AlP group was lower than that of control group. Also the levels of liver SOD in the AlP groups were less than control and in CDPI lower than alp group. Mean SOD levels of heart tissue were higher in CDPI group compared to ALP group. The mean GPx levels in CDPI group were lower than ALP group and in AlP group were lower than control group. NOx levels in heart tissue of AlP group were higher compared to control group Conclusion: Results of the present study showed that DPI has potential to reverse oxidative stress induced by aluminum phosphide exposure in rats associated with clinical improvement. It is proposed this project is repeated in more rat population. According to the drug is extensively studied, if the same results are achieved, it used for poisoned person.
