Study physiology of roots growth for soybean by WinRhizo pro-software with Vam3 genes

The mutant of Vam3 had great number of little vesicles substitute the big one of vacuole and disturbed ripening for the vacuolar proteins that also included alkaline phosphatase and carboxypeptidase Y. Vam3 implicated in the homotypic vacuolar fusibility. For that reason, our study presents here to identify the vam3 genes were studied to see the effect of root-scan by WinRhizo pro-software to analysis the roots with vam3 genes to see growth of roots after transgenic genes to soybean plants. The results here did not show any negative effect on root growth by Vam3 genes on soybean plants compared to control treatment. The study was including two different ways of root-scan. First one after transgenic genes to soybean plants with both overexpression and RNAi experiments. Results had shown good number of Length(cm), Surf- Area(cm2), Avg-Diam(mm), Root-Volume(cm3), Tips, Forks, and Crossings using winRHIZO Pro software for root growth compared to control in both tests overexpression and RNAi in twodifferent times after 30 and 60 days of planting. Gene Vam3-2 had shown good results to give higher positive effect to improve roots growth of root- scan Length(cm) was 2337.7878 compared to 806.1024 in the control at 60 days of planting. The number of root tips and forks were 584.8, 3398.6 respectively in Vam3-1 gene in the RNAi experiment after 60 days of planting in the greenhouse compared to 2221.2, 11433.8 respectively in the control with (pRAP17).
