Background and aims: Most chromosomal abnormalities in embryos are deadly and can causeembryonic growth to prevent before implantation or spontaneous abortion of the embryo duringthe first trimester of pregnancy. Using Morphological characteristics or a biopsy of a single cell ora small number of cells for genetic testing are typical methods to choose the best embryo. Thesemethods are Unreliable or invasive. Therefore, it is essential to detect normal embryos created byIVF or ICSI before transferring them to the uterus.Method: To establish the analysis protocol, ۳۰ spent human embryo culture medium samples(blastocyst) with defined morphologic grades were collected and measured using Raman spectroscopy.Individual Raman spectra of the embryo culture medium was analyzed to find biologicalcomponents. To validate the protocol via Principal Component Analysis, additional ۲۵۰ Ramanspectra from ۳۰ embryo culture media were analyzed.Results: Mean-centered Raman spectra and principal component analysis were differences inthe footprints of the highest-grade and the lowest-grade embryos growing in the culture medium.Stacking classification based on PCA achieved an overall accuracy of ۹۸% in correctly assigningeither based on Raman spectra.Conclusion: This study suggests that chromosomal abnormalities in embryos lead to changes inmetabolic footprints in embryo growth medium. This feature could be detected by Raman spectroscopyand, using machine learning, was analyzed by Raman- based footprint profiling of spentculture media.
