Among the cancers, breast cancer is the most common, accounting for ۲۳% of cancer diagnoses and ۱۴% of cancer deaths each year (۱). Nrf۲ is considered a double-edged sword as it plays a role in cancer prevention and development. Maintaining cellular redox homeostasis, anti-inflammatory activity, and inhibiting tumorigenesis are the mechanisms that correlate with carcinogen suppression while promoting cell proliferation, angiogenesis, self-renewal of cancer stem cells, and apoptosis suppression are the main mechanisms associated with cancer promotion and a poor prognosis through an excess expression of Nrf۲ (۲, ۳). Breast tumors with high expression of Nrf۲ have a worse prognostic and clinical outcome (۴). Moreover, constitutive activation of Nrf۲ can build up chemoresistance towards the chemotherapeutic drugs that have become a recent concern in managing cancer therapy (۵). The Nrf۲ pathway's potential therapeutic targets, as well as treatments and mechanisms that lessen chemoresistance in the treatment of breast cancer, are covered in this review.
