Effects of Different Oxygen Levels on the Spleen Histopathology in Two Weight-Groups of the Great Sturgeon (Huso huso)

In the present study, the influence of water oxygen concentration on the spleenhistopathology of juvenile great sturgeon (Huso huso) was evaluated in two weightgroups(initial weight 280.9 ± 49.2 g and 1217.9 ± 138.1 g). Oxygen treatments werehypoxia (2-3 mg/L), normoxia (5-6 mg/L) and hyperoxia (9-10 mg/L). Thespecimens were acclimated to the experimental tanks for one week, then, allocatedrandomly to 9 tanks in each initial weight groups (3 specimens per tank in higherinitial weight group and 6 fish per tank in lower initial weight group). The specimenswere kept for 8 week. To find the histopathological changes, the spleen of thespecimens were sampled. Histological samples were dehydrated using the routinemethods and embedded in paraffin wax, sectioned by a microtome and stained withH & E. In hypoxia treatment, both weight classes showed congestion. In lowerweight class, cell swelling, piknotic nuclei and loss of cytoplasm were observed. Inhyperoxia, both weight classes cell swelling and vacuolation of cytoplasm werefound. In both weight classes, there were significant differences in the weight of thespecimens and the spleen.
