Association between HLA-DRB1*01,HLA-A3,HLA-A24 alleles and Susceptibility to the Multiple Sclerosis disease in Isfahanian Patients with Multiple Sclerosis

Multiple sclerosis (MS) is a prototype of inflammatory demyelinating disease of the central nervous system and causes progressive disability in afflicted with MS. The etiology of MS remains unclear, however it is known as a multi factorial disease. It seems it is the result of interaction of genetic and environmental factors. Among all candidate genes responsible for the disease, MHC coding genes have the greatest effect on the risk of development of this disease. In particular, the gene of Human Leukocyte Antigen (HLA) which is located on chromosome 6p21 and is a part of the MHC region may contribute in the onset and progress of MS. MHC molecules, including class I and II, and their haplotypes have been studied well and seems to be associated susceptibility to MS disease.In this study, genomic DNA from peripheral blood samples of 89MS patients and 83 healthy controls were extracted and the association of HLA-A24, HLA-A*03 from HLA class I, and HLA-DRB1*1501 from HLA class II region was assessed by with susceptibility to MS by using PCR-SSP method.Statistical analysis using chi-square test and logistic regression, showed that there is an association between HLA-A24 allele and risk of MS (OR = 2.855, p-value = 0.001).On the other hand HLA-DRB1*1501 (OR = 0.749, p-value = 0.527) and HLA-A*03(OR = 0.46, p-value = 0.520) did not show a direct association with the progress of MS. Thus, we concluded that the presence of HLA-A24 may increase the risk of MS onset in Isfahanian population.
