چکیده مقاله:
Introduction: Chemerin is a novel adipocytokine that act in different tissues via its specific receptor, CMKLR1. it has been confirmed that over expression of chemerin and CMKLR1 are associated with cardiomyocyte apoptosis and cardiovascular diseases. The aim of this study was to investigate the effect of insulin resistance induced by high calorie diet on the gene expression pattern of chemerin and CMKLR1 in cardiac muscle of mice.Materials & Methods: Thirty male NMRI mice were devided to following groups: insulin resistant (n=15) and control (n=15) groups. insulin resistance was induced by administration of high fat/ high carbohydrate diet for 5 weeks and a single dose of streptozotocin (30 mg/kg; i.p). At days 7, 14, and 28 post treatment, the expression of chemerine and CMKLR1 in cardiac muscle were determined using real time PCR analysis. Glucose, insulin concentrations and HOMA-IR as insulin resistance markers were also determined using conventional methods. Results: Chemerin and CMKLR1 mRNA expression were upregulated in cardiac muscle, 14 or 28 days after diabetes induction, respectively. positive correlations were observed between chemerin and its receptor expression with levels of insulin, glucose and insulin resistance index, HOMA-IR.Conclusion: Due to adverse effect of chemerin on cardiomyocyte it was concluded that over expression of chemerin and its receptor in cardiac muscle of insulin resistance may had functional role on metabolic disturbances of cardiac muscle in insulin resistance state.
