Breast tumor kinase targeted therapy: challenges, biologic approach, and progress

چکیده مقاله:
Introduction and Aim: Kinases, which are a group of enzymes belonging to the class 2 enzyme(transferase). By transferring phosphate groups to various proteins, they play a role in regulatingthe broad metabolic pathways. Studies have shown that about 600 genes for kinases are humandoses of the genome, some of which play a role in the onset and spread of many cancers,including breast, and studies have identified the specific inhibitors of this enzyme; Smallmolecules kinase inhibitor has been shown to be effective in the treatment of certain cancers,including breast.Search Methods: This study was conducted through the review article on related topics inGoogle scholar, PubMed, Scopus web sites, and a searchable library of breast tumor, kinase,biological approach, challenge, progress keywords among numerous articles between 1993 and2019, and 67 articles related to the subject was collected and analyzed.Results: In general, inhibitors of kinases that have been confirmed by United States Food andDrug Administration, in some ways, inhibit the enzyme, which include: hydrophobic connectionswith active amino acids, interactions with the amino acid gate kiiper, interaction with aminoacids in the region Enzyme activation; also specific antibodies identified in breast cancer againstkinases including Trastuzum and T-DM1, which inhibit ERBB2 / HER2; and inhibitors of PI3Kand mTOR in breast cancer have been identified in Phase 3 clinical trial. Nowadays, extensivestudies focusing on the identification of small kinase inhibitors of natural nature are ongoing forthe treatment of breast cancer; for example, resveratrol inhibits the expression of ACT kinase inbreast cancer, but the variation in the kinase building expressed in Cancer cells have challengedthe design and manufacture of intruders; for example, Trastuzum did not yield the same results inbreast cancer treatment.Conclusion: The future of breast cancer treatment appears to be highly hopeful and growingbased on the targeting of kinases, and more extensive studies are required.
