Evaluation of rotenone effects on anxiety- and depression-like behaviors in Parkinson s male mice

Parkinson s disease (PD) is considered to be a movement disorder, and its diagnosis is based on the presence of a set of cardinal motor symptoms. Additional neuronal regions, are potentially involved in PD progression and maybe contribute to the appearance of non-motor symptoms. So the present study aims at providing new and more specific evidence for the validity of the rotenone model of PD by measuring the Anxiety and depression-like behavior in the male mice model of Parkinson s disease. Thirty male mice (30±2) were randomly divided into 3 groups; control group, rotenone solvent group, and rotenone group (ROT). ROT was treated with rotenone injections (2 mg/kg/48h) for 19 days. After comparing weight changes in the three groups, three behavioral tests (rotarod, open-field, and bar test) were run in order to check the development of Parkinson model. Then, the elevated plus maze and forced-swim test were run for measuring Anxiety and depression-like behavior in ROT. Rotenone treatment decreased the number of the entry in the open arms and the time spent on the open arms in the elevated plus-maze (P <0.05). As well as ROT were significantly more immobile during the forced-swimming test (P < 0.05). Rotenone could raise anxiety and depression-like behavior in the male mice model of Parkinson s disease. It seems that rotenone can affect other areas of the brain that are effective in anxiety and depression-like behavior
